Filaggrin (protein)

  • Filaggrin is a small protein which plays an essential function in the “mortar” allowing the wall made of keratinocytes (“bricks”) to be an effective barrier to the outside world.
  • In the Stratum Corneum of the Epidermis filaggrin exists as a precursor called profilaggrin.
    • In more detail…Located in granules within the cells (keratohyalin), the larger Profilaggrin protein (350Kd) undergo processing to yield individual filaggrin monomers at the transition between the stratum granulosum and the stratum corneum.
    • Within the stratum corneum, filaggrin monomers can be incorporated into the lipid envelope, which is responsible for the skin barrier function. Alternatively, these proteins can interact with keratin intermediate filaments.
    • Filaggrin undergoes further processing in the upper stratum corneum to release free amino acids that assist in water retention.
  • When Filaggrin is defective, individuals are strongly predisposed to Atopic Dermatitis, Ichthyosis Vulgaris, and dry Skin (xerosis).
